TREM-1–mediated pathways regulate Mcl-1 protein levels. (A) Adenovirus expressing TREM-1–infected THP-1 monocytic cells were cultured on TREM-1Ab– or phosphate-buffered saline–coated 24-well plate for 24 h, in combination with or without 0.6 μg/ml staurosporine for the last 4 h. Cells were collected and lysed for Western blot analysis with related antibodies against proapoptotic/antiapoptotic proteins and cytochrome c. The result is representative of three independent experiments. (B) Adenovirus expressing TREM-1–infected THP-1 monocytic cells were cultured with 50 mm PD98059 and/or 100 nm wortmannin for 1 h. Then, the cells were added to TREM-1Ab– or phosphate-buffered saline–coated 24-well plate for 24 h, in combination with or without 0.6 μg/ml staurosporine for the last 4 h. The Mcl-1 protein level in the cell lysate was detected using Western blot analysis. The result is representative of three independent experiments.
