Sevoflurane increased the interaction of postsynaptic density 95 protein (PSD-95) and ubiquitin E3 ligase mouse double mutant 2 homolog (MDM2). (A) Hippocampal neurons were treated with 4.1% sevoflurane for 4 h. The extracts of neuron were first subjected to immunoprecipitation (IP) using antibody against MDM2 and then were immunoblotted (IB) with PSD-95 antibody. The sevoflurane treatment induced a robust increase in the interaction of PSD-95 and MDM2 (lane 2 in upper panel) as compared to control conditions (lane 1 in upper panel). (B) In the total neuron extract, the sevoflurane treatment decreased total PSD-95 levels (lane 2 in upper panel) as compared to the control conditions (lane 1 in upper panel). There were equivalent amounts of β-actin in each condition (lower panel).
