Fig. 2.
MG132 attenuated the sevoflurane-induced reduction in postsynaptic density 95 protein (PSD-95) level in hippocampus neurons, synaptosomes of mice, and mice hippocampus tissues. (A) Anesthesia with 4.1% sevoflurane plus dimethyl sulfoxide (DMSO) for 4 h (lane 2) decreased PSD-95 levels as compared to control conditions plus DMSO (lane 1) in hippocampus neurons of mice. However, the treatment with sevoflurane plus MG132 (lane 4) did not reduce PSD-95 levels as compared to the treatment with control conditions plus MG132 (lane 3). (B) Quantification of the Western blot showed that sevoflurane plus DMSO (black bar) decreased PSD-95 levels as compared to control conditions plus DMSO (white bar). Sevoflurane plus MG132 (striped bar) did not decrease PSD-95 levels as compared to control conditions plus MG132 (gray bar; F = 11.030, P = 0.003, two-way analysis of variance [ANOVA]). (C) Anesthesia with 4.1% sevoflurane plus DMSO for 4 h (lane 2) decreased PSD-95 levels as compared to control conditions plus DMSO (lane 1) in hippocampus synaptosomes of mice. However, the treatment with sevoflurane plus MG132 (lane 4) did not reduce PSD-95 levels as compared to the treatment with control conditions plus MG132 (lane 3). (D) Quantification of the Western blot showed that sevoflurane plus DMSO (black bar) decreased PSD-95 levels as compared to control plus DMSO (white bar). Sevoflurane plus MG132 (striped bar) did not reduce PSD-95 levels as compared to control conditions plus MG132 (gray bar; F = 4.728, P = 0.042, two-way ANOVA). (E) Anesthesia with 3% sevoflurane 2 h daily for 3 days plus DMSO (postnatal day [P] 6, P7, and P8; lane 2) decreased PSD-95 levels as compared to control conditions plus DMSO (lane 1) in the hippocampus tissues of mice harvested at P8. However, the sevoflurane anesthesia plus MG132 (lane 4) did not significantly decrease PSD-95 levels as compared to the control conditions plus MG132 (lane 3). (F) Quantification of the Western blot showed that the sevoflurane anesthesia plus DMSO (black bar) decreased PSD-95 levels as compared to the control conditions plus DMSO (white bar). The sevoflurane anesthesia plus MG132 (striped bar) did not reduce PSD-95 levels as compared to control conditions plus MG132 (gray bar; F = 11.030, P = 0.003, two-way ANOVA). N = 6 in each group.
