Cultured cells were pretreated with 3 μm SP600125 or vehicle for 60 min, then stimulated by 0.1 mm midazolam or treated with vehicle (each for 20 min). Cell lysates were subjected to sodium dodecyl sulfate-polyacrylamide gel electrophoresis followed by Western blot analysis using antibodies against phosphospecific SAPK/JNK or SAPK/JNK. Similar results were obtained in two other cell preparations.
